These stresses are analyzed by imaging techniques (confocal microscopy, flow cytometry) (M2C Thierry Gauthier), molecular and cellular biology relating to signaling pathways (RT-qPCR, WB, SiRNA) coupled with cellular analyzes in real-time : video-imaging, impedance and measurements of oxygen consumption and acid efflux.


Particular attention is paid to metabolic reprogramming. Metabolic adaptability has a major role in the cellular defense to contaminants.
These aspects are addressed by physiological approaches, omics, including transcriptomic analyses (TRiX Yannick Lippi), metabolomics (AXIOM Cécile Canlet, Justine Bertrand-Michel I2MC, INSA Floriant Bellvert METATOUL) and modeling networks (E2 Nathalie Poupin and Fabien Jourdan), with the support of the BIOSTAT platform for data integration (Nathalie Villa-Vialaneix), as part of the METAhCOL project (ITMO Cancer Environment 2017-2020).


The in vivo mechanistic aspects  are set up on zebra-fish with different genotypes, in collaboration with UMS006 / CreFre (Genevière Tavernier). These models are obtained by crossing already existing lines and by tissue-specific invalidation by CRISPR-Cas9 technology for xenosensors. On the one hand, they will enable in vivo validation of the mechanisms of action and cellular responses demonstrated in vitro, using real-time imaging, histology and functional analysis techniques. On the other hand, as whole organisms, they will allow us to study more specifically carcinogenesis and aging.
